Neutralizing antibodies (NAb) have been identified as key molecules for the treatment, prevention and research of COVID-19 disease. However, due to the need to evaluate the neutralization performance of many candidates at an early stage, the development of NAb against pathogens can be a complicated process. Traditional methods of measuring virus neutralization include the use of the time-consuming virus neutralization test (VNT), pseudovirus neutralization test (pVNT), or table reduction neutralization test (PRNT), which requires also the use of facilities with strict pathogenic toxicity. (biosafety level 3). Regarding SAV-CoV-2 (the causative agent of COVID-19 disease), NAb is defined as the interaction between the virus-blocking spike protein (specifically its receptor binding domain (RBD)) and the Ability virus. This antibody along with other therapeutic antibodies were tested in surrogate virus neutralization test using spike trimers and ACE2 protein. This antibody showed expected neutralization of wild type, Alpha, Gamma, and Delta variants (IC50 24.23 ng/mL for the wild type, 86.18 ng/mL for Alpha, >500 ng/mL for Gamma and 8.44 ng/ml for Delta).
